I have a long layover at CLT — can I see anything?
With 5+ hours you can make a short Uptown run: the CATS Sprinter bus connects the airport to Uptown in roughly 25–35 minutes each way. Budget time for the security line coming back. Under 4 hours, stay put — the airport's rocking chairs are the famous consolation prize.
The Sprinter picks up outside baggage claim; check current fares and times at ridetransit.org before relying on it. If you do go, Romare Bearden Park and the Tryon Street museum block are the densest payoff within a few blocks of the transit center.

How do I get from the airport to Uptown?
Rideshare runs about 15 minutes to Uptown outside rush hour. The budget option is the CATS Sprinter bus (Route 5), which links CLT's terminal to Uptown — slower, but a few dollars instead of thirty.
There is no rail link to the airport. At rush hour, budget 30-40 minutes by road. Sprinter details and current fares: ridetransit.org.
If you have longer
A half day is enough for one district rather than a tour of the city. The Uptown wards are the obvious call from the airport bus; the Blue Line is the one ride worth taking for its own sake, though it does not reach the airport. Today knows what is actually open right now.
